Le présent travail concerne une étude phytochimique de deux plantes poussant dans les régions
semi-arides d’Algérie et appartenant à la famille Fabaceae : Astragalus monspessulanus L. et
Hedysarum carnosum DESF.. Les travaux d’extraction et de séparation par diverses méthodes
chromatographiques (CLV, CCM, CC, CLMP et CLHP), mis en oeuvre pour les deux espèces, nous
ont permis d’isoler pas moins de 25 métabolites secondaires appartenant principalement à deux
grandes classes de composés chimiques, que sont les composés phénoliques principalement
flavonoïdiques et triterpéniques. Leur identification a été réalisée grâce aux techniques d’analyses
spectroscopiques (RMN multi impulsionnelle et spectrométrie de masse), la mesure des pouvoirs
rotatoires et la comparaison avec les données de la littérature.
L’étude phytochimique portée sur les parties aériennes de l’espèce A. monspessulanus L. a
permis d’isoler et d’identifier 13 composés. Dix composés isolés de l’extrait n-butanolique se
répartissent en sept flavonoïdes (5 de type flavonols O-glycosylés, une chalcone et une aurone), un
lignane et deux saponines triterpéniques à squelette oléanane. La saponine connue sous le nom de
soyasaponine I est considérée comme marqueur chimiotaxonomique du genre Astragalus et de la
famille Fabaceae. Ainsi, trois composés ont été isolés de l’extrait acétate d’éthyle de cette espèce. Il
s’agit de deux phytostérols et un triterpénoїde. La composition chimique de l’espèce A.
monspessulanus L. est en parfait accord avec celle du genre Astragalus. En effet, ce travail a montré
encore une fois que les flavonoïdes et les saponosides triterpéniques sont les constituants dominants du
genre Astragalus.
Pour ce qui est de la plante Hedysarum carnosum DESF., 12 autres métabolites secondaires ont
été isolés de l’extrait acétate d’éthyle. Ils se répartissent en huit flavonoïdes (7 isoflavonoïdes et une
flavanone), un coumestane, un glycol, un dérivé d’acide benzoïque et un lignane. Ce travail montre
que les flavonoïdes, notamment les isoflavonoïdes, demeurent les constituants dominants du genre
Hedysarum.
L’extrait n-butanolique de l’espèce Astragalus monspessulanus L. a manifesté une activité
antioxydante modérée vis-à-vis du DPPH. Ce potentiel antioxydant pourrait être lié aux composés
phénoliques, particulièrement les flavonoïdes. En effet, les flavonoïdes sont considérés comme la
classe d’antioxydants la plus importante de la plante.
Il est à souligner que l'extrait acétate d’éthyle de Hedysarum carnosum DESF. n’a pas montré
une activité inhibitrice contre le radical DPPH. This work carried out the phytochemical study of two plants growing in semiarid regions of
Algeria and belonging to the family Fabaceae: Astragalus monspessulanus L. and Hedysarum
carnosum DESF. Our work of extraction and separation of the two species, by chromatographic
methods (VLC, TLC, CC, HMLC and HPLC), led to the isolation and characterization, by
spectroscopic methods (NMR, Mass spectrometry), measurement of the optical rotation []D and by
comparison with literature data, of 25 secondary metabolites belonging mainly to two big classes of
chemical compounds polyphenols and triterpenes.
The phytochemical study of the aerial parts of A. monspessulanus L. lead to the isolation and
characterization of 13 known compounds. Ten compounds were isolated from the n-butanol extract
including seven flavonoïds (5 with flavonols O-glycosides skelton, one chalcone and one aurone), one
lignan and two oleananes-type triterpene glycosides. The soyasaponine I oleanane is used as a
chemotaxonomic marker for the genus Astragalus. In addition, three secondary metabolites were
isolated from the ethyl acetate extract of this species including two phytosterols and one triterpenoid.
The chemical composition of the species A. monspessulanus L. is in full agreement with the plants of
the genus Astragalus. In fact, this study revealed that flavonoids and saponins are the major
constituents of this genus.
Twelve other known secondary metabolites were isolated from the ethyl acetate extract of the
second plant Hedysarum carnosum DESF. consisting of eight flavonoids, with 7 isoflavonoids and one
flavanon, one coumestan, one glycol, one benzoic acid derivative and one lignan. This study showed
that flavonoids, in particular isoflavonoids, remain the dominant constituents of the genus Hedysarum.
The n-butanol extract of Astragalus monspessulanus L. showed a moderate antioxidant activity
against DPPH. Thus, the antioxidant capacity is correlated to phenolic composition of the
corresponding extract, especially flavonoids. In fact, the flavonoid class is the most important plant
antioxidant. However, the ethyl acetate extract of Hedysarum carnosum DESF. did not show any
chelating activity of DPPH. يخص هذا العمل الدراسة الفيتوكيميائية لنبتين من الجزائر Hedysarum و Astragalus monspessulanus L.
و قد سمحت هذه الدراسة بعزل وتحديد بنية 25 مركب طبيعي و ذلك باستعمال .Fabaceae من عائلة carnosum DESF.
و باستعمال طرق التحليل الطيفي كمطيافية الرنين (HPLC, MPLC ,CLV, CCM, CC) الطرق الكروماتوغرافية
المغناطيسي النووي بجميع أنواعها، مطيافية الكتلة، حساب زاوية التدوير النوعي و المقارنة مع الدراسات السابقة. تنتمي
المركبات المعزولة إلى نوعين من الأيضات الثانوية الفينولية و التريتاربينية.
10 مركبات Astragalus monspessulanus L. تم عزل 13 مركب طبيعي معروف من الأجزاء الهوائية للنبتة
منها 7 فلافونويدات (5 منها ذات هيكل فلافونول جليكوزيلي، شالكون واحد و أورون واحد(، n-BuOH من مستخلص
يعتبرمن المركبات المحددة للصنف soyasaponine I لينيان و 2 صاابونينات ذات هيكل أوليانان. الأوليان المعروف باسم
منها 2 فيتوستيرول و éthyle acétate بالإضافة إلى ذلك، 3 مركبات أخرى تم عزلها من مستخلص .Astragalus
تريتاربان واحد. هذا ما يتفق مع الدراسات السابقة لنباتات هذا الجنس اتفاقا تاما، حيث أن الفلافونويدات و الصابونينات تعتبر
المكونات النموذجية لهذا الجنس.
فقد تم عزل 12 مركب طبيعي آخر معروف من المستخلص Hedysarum carnosum DESF. بالنسبة للنبتة
منها 8 فلافونويدات تتضمن 7 أيزوفلافونويد و فلافا نون ، كومستان ، جليكول ، مشتق حمض بنزويكي ، éthyle acétate
.Hedysarum و لينيان. تبين هذه الدراسة أن الفلافونويدات، وبالأخص الإيزوفلافونويدات، تبقى المكونات الغالبة للجنس
فعالية ضد أكسدة معتدلة اتجاه Astragalus monspessulanus L. للنبتة n-BuOH لقد أظهر مستخلص
هذه الفعالية راجعة إلى المركبات الفينولية لهذا المستخلص، خاصة الفلافونويدات. في الحقيقة، تعتبر الفلافونويدات .DPPH
Hedysarum للنبتة éthyle acétate من أهم المواد المضادة للأكسدة في النباتات. في حين لم يظهر المستخلص
.DPPH أي فعالية مثبطة اتجاه carnosum DESF. |
