dc.description.abstractCe présent travail de recherche a porté sur l’investigation phytochimique d’une plante nommée Ononis mitissima L., appartenant à la famille Fabaceae. Il a été consacré également à une évaluation biologique à travers les activités biologique, anti-oxydante, anti-hémolytique et anti-inflammatoire de ses extraits (éther de pétrole, acétate d'éthyle et n-butanol). L’étude phytochimique réalisée sur les extraits AcOEt et n-BuOH a permis d’isoler et d’identifier 14 métabolites secondaires. Le processus de séparation de ces composés s’est basé sur les méthodes chromatographiques (VLC, CCM et CC) et l’élucidation structurale par les méthodes modernes d’analyse spectroscopiques (RMN multiimpulsionnelle, la spectrométrie de masse à haute résolution HR-ESI-MS, la mesure du pouvoir rotatoire [α]D20 et par la comparaison avec les données de la littérature. L’extrait AcOEt de l’espèce Ononis mitissima a fourni dix composés se répartissant en un isoflavonoïde, deux flavones, un ptérocarpane, un dérivé du résorcinol, deux phytostérols et trois triterpènes, dont un à squelette lanostane s’est avéré original. A partir de l’extrait butanolique, 4 métabolites secondaires ont été identifiés. Il s’agit d’une saponine triterpénique à squelette oléanane, deux isoflavonoïdes glucosylés dont un à groupement 3-hydroxy-3-methylglutaryle nouveau et un flavonol 3-glycosilé. La composition chimique de l’espèce étudiée est en parfait accord avec celle des plantes du genre Ononis. En effet, ce travail de recherche a montré encore une fois que les flavonoïdes sont les constituants majoritaires du genre. Les résultats du dosage des polyphénols totaux et flavonoïdes des extraits EP, AcOEt et n-BuOH de la plante Ononis mitissima, obtenus par les méthodes de Folin-Ciocalteu et de trichlorure d’aluminium (AlCl3), révèlent que l’extrait AcOEt est le plus riche en polyphénols totaux et flavonoïdes par rapport aux extraits EP et n-BuOH. Les résultats de l'activité anti-oxydante par piégeage des radicaux DPPH, pouvoir réducteur de fer et capacité antioxydante totale par le phosphomolybdate ont montré que l'extrait AcOEt présente le plus grand pouvoir par rapport aux extraits EP et n-BuOH. L'extrait EP a révélé un pourcentage d'inhibition plus élevé (85,26%) que l'acide ascorbique en standard (62,36%) à la concentration de 100 μg/mL dans le test de piégeage du peroxyde d'hydrogène et une capacité antioxydante plus élevée (42,07 ± 0,25%) que le BHA (41,92 ± 0,44%) dans le dosage du thiocyanate ferrique. Tous les extraits ont montré une activité anti-inflammatoire modérée avec des pourcentages d'inhibition respectifs de 21,96, 33,41 et 11,48 %, par rapport au diclofénac (86,72%), à la même concentration. L'activité anti-hémolytique la plus forte a été observée avec l'extrait AcOEt dans une IC50 égale à 0,65 mg/ml. This work is focused on the phytochemical investigation of a plant called Ononis mitissima L., belonging to the Fabaceae family. It also concerning of a biological evaluation through the biological activities, anti-oxidant, anti-hemolytic and anti-inflammatory of its extracts (petroleum ether, ethyl acetate and n-butanol). The phytochemical study carried out on EtOAc and n-BuOH extracts allowed the isolation and identification of 14 secondary metabolites. The separation process for these compounds was based on chromatographic methods (VLC, TLC and CC) and structural elucidation by modern methods of spectroscopic analysis (NMR, high resolution mass spectrometry HR-ESI MS, measurement of the optical rotation [α]D20 and by comparison with data from the literature. The EtOAc extract of the species Ononis mitissima provided ten compounds consisting of an isoflavon, two flavons, a pterocarpan, a resorcinol derivative, two phytosterols and three triterpenes, from which one new triterpene with lanostane skeleton. From the butanol extract, 4 secondary metabolites have been identified. These compounds consisted of a triterpenic saponin with oleanane skeleton, two isoflavon glucosides, from which one new has a 3-hydroxy-3-methylglutaryl group and a flavonol 3-glycosid. The chemical composition of studied species is in full agreement with the plants of the genus Ononis. Indeed, this research work has shown once again that flavonoids are the major constituents of this genus. The results of total phenolic and flavonoid contents of the EtOAc and n-BuOH extracts from species Ononis mitissima obtained by the Folin-Ciocalteu and AlCl3 methods revealed that EtOAc recorded the highest content of polyphenols and flavonoids compared to the PE and n-BuOH extracts. The results of the antioxidant activity by DPPH radical scavenging activity, ferric reducing power and total antioxidant capacity by the phosphomolybdate showed that the EtOAc extract has the greatest power compared to the PE and n-BuOH extracts. The PE extract revealed a higher percentage of inhibition (85.26 %) than ascorbic acid as standard (62.36 %) at the concentration of 100 μg/mL in hydrogen peroxide scavenging test and a higher antioxidant capacity (42.07 ± 0.25 %) in the ferric thiocyanate assay compared to the BHA (41.92 ± 0.44 %) as a reference. All the tested extracts had moderate anti-inflammatory activity with the percentages of inhibition at 21.96, 33.41 and 11.48 % respectively, compared to diclofenac (86,72 %). The strongest anti-hemolytic activity was observed in EtOAc extract with a value of IC50 at 0.65 mg/mL. ركز هذا البحث على الدراسة الفيتو كيميائية للنبتة Ononis mitissima L. ، التي تنتمي إلى عائلة البقوليات. كذلك تمت الدراسة البيولوجية من خلال تقييم النشاط المضادة للأكسدة، التأثير المضاد للالتهاب و النشاط المضاد لانحلال الدم للمستخلصات العضوية (أثير البترول، الأستات الإيثيلي و البوتانول) . أدت الدراسة الفيتوكيميائية لمستخلصي الاستات الايثيلي و البوتانول إلى عزل وتحديد 14 مركب طبيعي. استندت عملية فصل هذه المركبات إلى طرق كروماتوجرافية (CC و CCM و VLC) . اعتمد التوضيح الهيكلي لهذه المركبات على الطرق الحديثة للتحليل الطيفي (مطيافية الرنين النووي المغناطيسي بجميع انواعه، مطيافة الكتلة، حساب زاوية التدوير النوعي [α]D20 والمقارنة مع الدراسات السابقة . أعطى مستخلص الأستات الإيثيل لنبات Ononis mitissima 10 مركبات مقسمة إلى 1 إيزوفلافونويد، 2 فلافونون، 1 بتيروكاربان، 1 مشتق ريسورسينول، اثنين من فيتوستيرولات و ثلاث مركبات ثلاثية التربينات، أحدها مركب جديد ذو هيكل لانوستان . انطلاقا من مستخلص البيوتانول، تم تحديد 4 مركبات. وهي عبارة عن صابونين ذو هيكل أوليانان، و اثنين من إيزوفلافونويدات غلوكوزيلاتية، أحدهما جديد يحتوي على مجموعة ميثيل جلوتاريل و فلافونول- 3 جليكوزيلاتي . التركيب الكيميائي للنوع المدروس يتفق تمامًا مع الدراسات السابقة لنباتات الجنس Ononis حيث أظهر هذا البحث مرة أخرى أن الفلافونويدات هي المكونات الرئيسية لهذا للجنس . تكشف نتائج المحتوى الإجمالي لمتعددات الفينول و الفلافونويدات لمستخلصي الاستات الايثيلي و البوتانول لنبات Ononis mitissima بواسطة طريقتي Ciocalteu-Folin و ثلاثي كلور الألمنيوم (AlCl3) على التوالي أن مستخلص الاستات الايثيلي هو الأغنى بالمواد البوليفينولية و الفلافونويدية مقارنة مع مستخلصي البوتانول و الأثير البترولي. أظهرت نتائج النشاط المضاد للأكسدة عن طريق النشاط الازاحي للجذر الحر DPPH ، القوة المضادة لأكسدة الحديد (FRAP) واجمالي القدرة المضادة للأكسدة عن طريق الفوسفوموليبدات (PPM) ، أن مستخلص الاستات الايثيلي يبدي أكبر قوة مقارنة مع مستخلصي البوتانول و الاثير البترولي. كما أظهر مستخلص الاثير البترولي نسبة تثبيط أعلى (85.26 %) من حمض الأسكوربيك كدواء مرجعي (62.36 %) بتركيز 100 ميكروغرام/مل في النشاط المضاد للأكسدة عن النشاط الازاحي ل H2O2 وقدرة مضادة للأكسدة ( %0.25± 42.07 )أعلى من BHA (41.92 ± 0.44 %) في معايرة ثيوسيانات الحديد. أظهرت جميع المستخلصات نشاطا معتدلاً مضاداً للالتهاب بنسب 21,96 % ، % 33.14 و 11.48 % مقارنة بالديكلوفيناك (86.72 %) وبنفس التركيز. سجل مستخلص الاستات الايثيلي أقوى نشاط مضاد لانحلال الدم بتركيز IC50 يقدر ب 0.65 مغ/مل .fr_FR
